With increasing age human males are likely to develop benign prostatic hyperplasia (BPH), a disorder characterized by urethral obstruction due to an increase in size of the prostate gland. Drug treatments of this condition are not entirely satisfactory and the current project is to examine the mechanisms by which the prostate grows and occludes the urethra. The incidence of cardiovascular diseases is much lower in women before menopause, and this is thought to be due to a beneficial effect of oestrogen on the cardiovascular system. However the mechanisms of the hormone’s cardiovascular-protective actions are still not clear. The proposed project will determine whether oestrogen acts by slowing the natural ageing process of cells.

TNF-related apoptosis-inducing ligand (TRAIL) is a protein with potentially useful actions in human health and disease. TRAIL is able to prevent atherosclerosis, the cause of heart attacks and strokes. In addition, we have recently shown that its actions on fat and the pancreas may prevent the development of the metabolic syndrome and type 2 diabetes. These studies will explore the therapeutic potential of TRAIL for the prevention of diabetes and heart disease in a range of animal models.
